Is it possible to back-up just an application? i.e. garageband? I don't won't back-up the entire hard drive just an application. The software has to copy all associated files also.

An application such as Garageband is available on your restore disks, and you can always reinstall from those disks. Garageband is probably not a good example, as it has files stored in a variety of places on your hard drive.
Most other applications are self-contained in their application package file (actually a type of folder), so you can just copy those off to another drive.
If you also need the files created by those applications, then that will depend on where you chose to store those files, such as the iTunes music folder, for example.
Would be a help if you could tell why you need to backup an application that is fairly easy to reinstall from your original disks, where you already have a backup of your apps....

The reason I was interested in Garage was this. I upgraded form 10.4 to 10.5. Garageband was included in 10.4. When I upgraded from 10.4 to 10.5, the 10.5 install disk said it can't install on this computer, and it gave my the option to do a clean install and erase my entire hard drive. Since all my program files, i.e. illustrator, protools, etc. I was not concerned. When the install was done, garageband was gone, Can I reinstall Garageband from 10.4 with messing up 10.5?

GarageBand is not part of any OS X install. It _is_ on your original restore disks, as part of the bundled software, and can be custom installed with the older disk.
