Ran the Software Update:
2007-07-02: Installed "Pro Application Support" (4.0)
2007-07-02: Installed "Pages Update" (2.0.2)
2007-07-02: Installed "Keynote Update" (3.0.2)
2007-07-02: Installed "iPhoto Update" (6.0.6)
2007-07-02: Installed "QuickTime" (7.1.6)
2007-07-02: Installed "iTunes" (7.3)
2007-07-02: Installed "MacBook Pro Software Update" (1.0)
2007-07-02: Installed "Security Update 2007-006 (Universal)" (1.0)

Entourage won't launch following update and neither will the Office Setup Assistant on the installer CD.

Word launched okay still.

Checked through exactly what preceded the fault. Had also made several (what I thought were harmless) changes to the system settings using the Mac Pilot utility downloaded from the Apple Downloads site. Tried to reset all of these to defaults but to no avail. Not exactly certain of the sequence now, but this could be related.
Anyway, ended up re-installing Mac OSX from the original discs using "Archive and Install" method to save as many settings as possible. This resolved the issue, although had to follow up by re-installing several other applications.
Ran the Software Update again but excluded the latest "Security Update" and the "MacBook Pro Software Update"
Seems to be working okay now.
Not quite game to let these run just yet.

After running all those updates, did you open Disk Utility and Repair Permissions?

Hi Cheryl,
Yes I did. The disk verify and repair found errors but were unable to make them good. I also ran the repair permissions after I re-installed the system several time just to be sure.

When the updates were installing, were you using the computer at the same time? There is a trick that I found after experiencing trouble after an update.

Watch the install screen in Software Update. It tells you what the installer is doing - i.e. download, verify, writing, and most important - optimize. When it is optimizing system - stop using the computer. This includes any games, email or internet surfing. The installer checks all the files and if one is being used, it will cause some strange problems.

When Disk Utility can not make repairs, it is time for a third party utility. Disk Warrior or TechTool Pro will do the job. There is no need to install the program. Just use the CD to start up the computer and allow it to repair the hard drive.

Thanks for your reply. I didn't notice, but I could easily have been using this while Software Update was in the optimising phase. I'll definitely keep an eye on this in future.

I have Disk Warrior too but I didn't use it on this occassion - should have thought of that myself - doh!
